Beckman-Coulter EPICS XL-MCL.
Our facility have 3 Beckman-Coulter EPICS XL flow cytometers. They use an argon ion air cooled laser (emission at 488nm/15mW power), both have 5 PMT detectors for 90LS and 4 color detection and 1 diode detector for FLS.

We have recently installed our fourth analyzer, a Beckman-Coulter FC-500. This instrument has a 488nm argon laser and a 637nm HeNe laser and can simultaneously measure up to 5 color parameters.
Parameter Spectra Fluorochrome
FLS 488nm N/A
SS 488nm N/A
FL1 525nm FITC , GFP, YFP, CFSE,
FL2 575nm PE, DsRed, PI
FL3 610nm PETxRd, PI, Alexa610
FL4 675nm PECy5, APC, 7AAD, PerCP
FL5 750nm PECy7, APC-Cy7
Note: APC and APC-Cy7 are excited by the 637nm HeNe Laser.
Each instrument has its own computer workstation consisting of a Pentium 120 PC. We currently use System II Data Acquisition and Display software, version 2.0 . This program works in a DOS Platform integrated with a Windows 95/98 operating system environment.
Beckman-Coulter EPICS Elite ESP Sorter
Cell sorter equipped with an argon ion 15mW air cooled laser, a He-Ne 15mW air cooled laser, and a water cooled 5W argon ion laser. The Elite sorter also has a Gated Amplifier and an Autoclone for sorting individual cells into a microtiter plate, and 4 fluorescence detectors. This instrument is capable of sorting at data rates approaching 5,000 events per second (18 million events per hour). Sorting time has to be arranged with the facility’s operator. It is not unusual to schedule sort time 2 weeks in advance. Investigators using the Elite cell sorter are required to provide 2.5 L of sterile PBS for sheath. In addition, as with all flow cytometry samples, you have to filter your sample through a 40um nylon mesh to remove large clumps of cells and debris.
DAKO-Cytomation MoFlo High Speed Sorter
Equipped with 2 Coherent Innova I-90 (argon ion gas, tuned for 488nm and UV excitation) water cooled lasers and 1 Coherent Innova I-70 Spectrum laser tuned for 647nm excitation. This instrument is capable of sorting at data rates exceeding 20,000 events per second (72 million events per hour). Other specifications include:
-7 Fluorescent detectors
-Cyclone sorting (single cell deposition to microtiter plates)
-4 way sorting
Other Services: We also have a Pentium IV, DELL computer with a HP laser printer. This PC has EXPOII, and WinMIDI for analysis of listmode flow cytometry data, and MCycle from Phoenix Flow Systems for cell cycle analysis.
In addition, each cytometer workstation have the following; Iomega 2.0 GB JAZ and ZIP 250mb disk drives and CD Burners for data downloading.
